Reflection Electron Microscopic(REM) and Fluorescent Microscopic(LSM) Study about the Change of Bone Tissue around the Vent of IMZ Implant Implanted in Rabbit¢¥ s Tibia


Abstract
The purpose of this study was to observe morphologically bone formation and osseointegration of undecalcified bone, and peripheral tissue around the vent of IMZ imptants with 4. O§® in diameter and 8§® in length, being implanted in rabbit¢¥s tibia. After muscular injection of 25 §·/§¸ oxytetracyclin and alizalin red(20 §·/§¸) was done before 1 week and 3 weeks of sacrification, respectively in 3,6,8 months, the explanted IMT impants associated with bone tissue were fxed with 10% neutral formalin, dehydrated, and embedded with Spurr low viscosity resin. After the specimen was crossly cut by 30§­ with slow diamond wheel saw these were grinded up to 100§­ with grinder, and examined with light and laser scanning microscopy, For reflection eleorone microscopy, these specimens were coated by carbon. The obtained results were as follows.
1. In toluidin blue staining of 3 months, unilateral trabecular formatiom was attached to the natural bone. In reflection microscopy, there was a different bone density from natural bone and darkness in the portion of new bone foramtion. In LSM features, AZ and TC labelled new bone around the implant were seen prominendy.
2. In toluidin blue staining of 6 months, compact bone attached to the implant interface and new bone formation in the vent were seen around. In reflection microscopy, similar bone density were seen around the implant interface, but low density bone were seen in the vent. In LSM features, the distance between AZ and TC labelled bone adjacent to implant interface were prominently decreased.
3. In toluidin blue staining of 8 months, almost area of implant interface was attached with mineralized tissue and bone trabecular have transfered opposite site. Also seen isolated new bone islands. In reflection microscopy, calcified bone showed similar bone density to that of natural bone, but there was more bone formation toward the central portion than 6 months. In LSM features, AZ and TC labelled bone adjacent to the implant interface were rarely seen especially TC labelled bone was seen poorly, but in osteon of natural bone was double labelle by TC and AZ.
These results suggested that there were no complete bone formation in the vent at 8 months experimental group. But well calcified bone similar with the naural bone were seen around the implant interface. New bone formation aruund the implant interface had decreased at 6 months experimental group. Natural bone remodelling were still seen in 8 months experimental group.
